چکیده

The growth characteristics of an oncogenic simian adenovirus, SV20, in rhesus monkey (RhMK) and hamster kidney (HamK) cell cultures were compared. The ad sorption rate of SV20 in both cell systems was rather slow, but increased steadily up to 6 hr. There was 3 to 8 times more cell-associated virus in RhMK cells than in HamK cells during the first 6 hr of infection. A complete growth cycle of SV20 was obtained in infected monkey cells. This includes sequential nuclear alterations, pro duction of T antigen, viral antigen, and infectious virus. In hamster cells, SV20 induced an abortive infection. Eosinophilic inclusions and T antigen were observed as two distinct entities in the infected hamster cells, but no viral antigen or infectious virus was found in the hamster cell system. Marked stimulation of DNA synthesis in SV20-infected hamster cells was observed.

Serological studies of the T and tumor antigens of the oncogenic simian adenoviruses.

Tumor-specific antigens and antisera were prepared for eight of the oncogenic simian adenoviruses. Complement-fixation tests revealed three distinct serological subgroups. This grouping was maintained in studies of virus-infected cells (T antigens) although high titered preparations were obtained for only the major subgroup I. The current grouping is as follows: (I) SV1, SV11, SV25, SV33, SV34,...

Synthesis of the Ad2+ND5-specified 42K protein is regulated posttranscriptionally in abortively infected monkey cells.

In abortive infections of monkey cells by Ad2+ND5, the synthesis of the simian virus 40-specific 42,000-molecular-weight (42K) protein was reduced approximately 10-fold compared with a productive coinfection by Ad2+ND5 plus Ad2hr400 and about 20-fold compared with productive infections by Ad2+ND5 plus simian virus 40 or by Ad2+ND2 alone. However, the level of Ad2+ND5-specific mRNA was depressed...

Designing E1 Deleted Adenoviral Vector by Homologous Recombination

Adenoviruses are used extensively to deliver genes into mammalian cells, particularly where there is a requirement for high-level expression of transgene products in cultured cells, or for use as recombinant viral vaccines or in gene therapy. In spite of their usefulness, the construction of adenoviral vectors (AdV) is a cumbersome and lengthy process that is not readily amenable to the generat...

A murine teratocarcinoma stem cell line carries suppressed oncogenic virus genomes

Murine teratocarcinoma stem cells are nonpermissive for productive infection by a variety of DNA (polyoma and SV40 virus) and RNA (murine leukemia and sarcoma virus) tumor viruses whereas differentiated murine cells derived from the stem cells are permissive for productive (or abortive in the case of SV40) infection by these same viruses. The block to productive infection by these oncogenic vir...
